The Construction and the Determination
of the Analytical Characteristics of a Potentiometrical Biosensor for Glucose


Abstract:

The aim of this paper is to study the characteristics and performances of a potentiometric enzyme electrode for the analysis of glucose; glass electrodes were used for the preparation of enzyme-pH electrodes, by entrapping the enzyme (glucose oxidase) on the sensitive bulb of the glass electrode: the glucose oxidase was immobilized as a liquid layer trapped within a nylon membrane. Different types of nylon membranes were used, in order to obtain the enzyme electrodes: Biodyne A (nylon, 0,45 ìm porosity), Biodyne A (nylon, 1,21 ìm porosity ), Biodyne B (nylon positively charged, with -NH2 surface active groups, 0,45 ìm porosity) and Biodyne C (nylon negatively charged, with -COOH surface active groups, 0,45 ìm porosity). The enzyme, glucose oxidase, catalyses the oxidation of glucose to gluconic acid; the generation of the latter produces a pH decrease, that is proportional to the concentration of glucose; the pH decrease in time, for different glucose concentrations, was followed; then, calibration curves for the glucose oxidase - pH electrodes were obtained. The developed glucose enzyme electrodes allow the determination of glucose within 0.0001-0.05 M concentrations; the influence of enzyme loading, membrane porosity and membrane surface groups on the performances of the biosenzor (sensitivity, liniarity, detection limit) were investigated. The enzyme electrodes were applied to glucose analysis in orange juices (commercial products). The results were in accordance with those obtained by conventional methods.
