The Kinetics of the Hydrolysis Reaction of Phosphatidylcholine
in the Presence of A2 Phospholiphase Biosynthesized by Fusobacterium necrophorum


Abstract:

A protein presenting an A2 phospholipasic activity has been isolated from Fusobacterium necrophorum supernatant. This paper discusses the results provided by the protein purification and investigation on phospholipase A2 kinetic properties. For the purification of the protein, precipitation with ammonium sulfate, delipidation with ethylic ether and acetone, chromatography separation on Sephadex G 200 and DEAE cellulose have been carried out. Kinetic features of phosphatidylcholine hydrolysis were studied with egg yolk phosphatidylcholine emulsified in Tris HCl buffer at pH 7.5 (Triton X 100 and calcium ions have been added at concentrations of 0,1% and 10 mM, respectively).
